The Prostaglandin I2 Release of Seeded Endothelial Cells of Umbilical Vein on PTFE




Abstract
The endothelium serves as an important modulator of vascular homeostases by secreting various levels of both thrombotic and antithrombotic and antithrombotic agents. One of the important product of endothelial cells, prostaglandin I2 or
prostacyclin(PGI2) helps to maintain hemostasis through its involvement in coagulation, platelet activation, leukocyte migration and adhesion, vascular tone regulation and growth control.
The small diameter synthetic vaxscular grafts available today are inferior to native arteries and veins. There are many causes for failures of vascualar reconstruction, but important factor is the thrombogenic properties of the graft material
itself.
Endothelial cell seeding was attempted to improve the patency by investigators because synthetic material itself does not have intima of endothelial cells and antithrombotic properties.
This study was undertaken to investigate the capacity of not only cultured endothelial cells in media, but the seeded grafts to produce PGI2.
Endothelial cells were isolated from human umbilical vein and cultured in M199 medium plus 20% fetal calf serum. Purity of culture was determined by immunological fluorescent staining of factor¥·related antigen, phase-contrast-and scanning
electron
microscopy. TRK 790 radio-immunoassay kit was used for the measuring of 6-keto-PGF1¥áreleased by endothelial cells.
@ES The results were as follows:
@EN 1) The concentration of PGI2 released from the cultrued endothelial cells(1¡¿106) and seeded cells were 546.1¡¾41.3 pg and 548.91¡¾39.9 pg repectively.
2) Inhibition of endothelial cells with ibuprofen(20 umol/L) resulted in the decrease of PGI2 release (10.27¡¾1.0 pg in cultured cells and 9.84¡¾1.2 pg in seeded cells).
In conclusion, our data showed that the level of PGI2 released from the seeded endothelial cells were initially not different from that of cultured endothelial cells.
